Light and electron microscopic observations on the synovial membrane in carrageenan-induced arthritis of rabbit

Abstract
The early morphologic changes of knee joints were investigated in the carrageenan (CAR) -induced synovitis.
The amount of 1ml of 2 % crude CAR was injected into the joint via suprapatellar pouch in adult male rabbits, and they were sacrificed at various intervals up to seventh days. Light and electron microscopic examinations of knee joints were performed.
The results were obtained as follows.
1. The histologic changes in the synovial tissue showed early hemodynamic changes, multistratification with villous hyperplasia of synovial lining cells, proliteration of CAR-laden macrophages with its giant cell formation, infiltration of mononuclear cells, destruction of subsynovial adipose tissue and lysine of CAR-laden macrophages.
2. The early fine structural changes in the CAR-induced synovitis showed endocytotic activity of type A cells (vacuolar synovial cells: VSC) such as intracytoplasmic migration of vesicles, vacuoles, uptake and degradation of CAR in lysosomes, disintegration and heterolysis of lysosomes, and formation of residual body.
These results suggested that the CAR-induced synovitis might be induced by secondary lysosomal damage in type A synovial cells.
